Significance Statement:
- While it is known that two classes of actin nucleators, Arp2/3 and formins, regulate F-actin assembly, it remains unclear how their activities are coordinated to carry out diverse cellular functions.
- This study examines cortical actin waves in mast cells and finds that formin and Arp2/3-dependent nucleation of actin filaments is controlled by competing interactions with their shared regulator Cdc42.
- This finding challenges the prevailing model that Arp2/3 and formins compete for a limited pool of monomeric actin and provides new insights into the mechanisms of cortical actin wave regulation.
