Primary cilia are required for Hedgehog signaling. But why they are required is unknown. Francesc Garcia-Gonzalo, Siew Cheng Phua, and Jeremy Reiter postulated that Hedgehog signaling has a unique phospholipid platform for signaling. They found that disruption of Inpp5e, a phosphoinositide phosphatase, increased ciliary PI(4,5)P2, decreased Hedgehog signaling, and increased the ciliary localization of Hedgehog inhibitor Tulp3.
